Isolation, Screening and Extraction of Yellow Pigment Producing Bacterial Isolate from Shivnath River, Durg District, Chhattisgarh

Abstract

An unidentified yellow bacterial strain isolated from water from Shivnath river, Dist- Durg,
Chhattisgarh, it characterization step was done with performing morphological, microscopic,
biochemical identification as Bacillus sp. Small yellow pigment producing colony was measuring 1-
2mm in diameter on Luria Bertani agar medium was the striking feature noted in the organism. The
bacterium could grow over a pH 7.0 and temperature (37C) but optimal growth and pigmentation
was observed in LB medium containing agro-waste (rice bran powder) under submerged
fermentation. Scanning absorbance spectrum was analyzed (?max) by using UV-VIS
spectrophotometric at 200-800nm range. The optimum absorbance spectrum of yellow pigment
producing Bacillus sp. was 450nm it revealed the presence of carotenoid pigment in the bacterial
culture.
